NONMEM® is a non-linear mixed effects modeling software tool used in population pharmacokinetic- pharmacodynamic analysis. This section explains the procedure to develop a population PK or PKPD model for the problem given in case studies.

We have assumed the following things before any user uses this section:

o NONMEM V is installed on the PC.

o Since the tutorial section uses Wings for NONMEM, it is recommended to work using Wings for NONMEM and it can be downloaded from http://wfn.sourceforge.net/

o FORTRAN compiler is installed on the PC.

A NONMEM run requires the following

o Data file : User supplied text file containing all data for analysis. The file is created with a ".csv" or ".prn" extension.

o Control stream : User-written and supplied modeling instruction file for NONMEM. The file is created with a ".ctl" extension using any text editor.

Modeling process

It is assumed that the following two steps are performed before proceeding to modeling.

o The project plan is specified apriori.

o Initial preliminary data analysis has been performed and rough initial estimates are obtained.
